Assay methodology

  1. Antibody Binding: Two oligonucleotide-labeled antibodies bind to the same target protein in close proximity.
  2. Proximity Extension & DNA Polymerization: If both antibodies are bound, their DNA tags hybridize and are extended by DNA polymerase, creating a unique sequence.
  3. qPCR or NGS-Based Detection: The DNA sequence is amplified and quantified using real-time PCR (qPCR).
  4. Data Analysis: Results are processed using Olink Insight Software, offering detailed proteomic insights that can be integrated into statistical and bioinformatics tools.
